Cyanophycin Granules from the Blue-Green Alga Anabaena cylindrica: A Reserve Material Consisting of Copolymers of Aspartic Acid and Arginine.
نویسنده
چکیده
The cyanophycin granules from two-week-old cultures of the blue-green alga Anabaena cylindrica were isolated by sonication of the cells followed by differential centrifugation. These particles are composed of polypeptides containing only two amino acids, aspartic acid and arginine, and have a molecular weight between about 25,000 and 100,000.

Measurement of the cyanophycin granule polypeptide contained in the blue-green alga Anabaena cylindrica.
An assay was developed to determine the amount of cyanophycin granules in blue-green algae. The amount of this polypeptide in cells of Anabaena cylindrica was measured as a function of culture age and was compared with changes in other proteinaceous cellular components. The data presented support the notion that the cyanophycin granule is a cellular nitrogen reserve.
متن کاملCompartmentalized cyanophycin metabolism in the diazotrophic filaments of a heterocyst-forming cyanobacterium.
Heterocyst-forming cyanobacteria are multicellular organisms in which growth requires the activity of two metabolically interdependent cell types, the vegetative cells that perform oxygenic photosynthesis and the dinitrogen-fixing heterocysts. Vegetative cells provide the heterocysts with reduced carbon, and heterocysts provide the vegetative cells with fixed nitrogen. Heterocysts conspicuously...
متن کاملThe Fine Structure of the Nuclear Material of a Blue-green Alga, Anabaena Cylindrica Lemm
The chromatinic material of the blue-green alga Anabaena cylindrica has complex configurations in the central regions of the cells. The distribution of the chromatin within the cells varies in different filaments, probably in response to variations in the disposition of other cellular components. In electron micrographs of thin sections of organisms fixed by the method of Kellenberger, Ryter, a...
متن کاملCyanophycin synthetase-like enzymes of non-cyanobacterial eubacteria: characterization of the polymer produced by a recombinant synthetase of Desulfitobacterium hafniense.
Some bacterial genomes were found to contain genes encoding putative proteins with considerable sequence homology to cyanophycin synthetase CphA of cyanobacteria. Such a gene from the Gram-positive, spore-forming anaerobe Desulfitobacterium hafniense was cloned. Expression in Escherichia coli resulted in the formation of a polydispers copolymer of aspartic acid and arginine, with a minor amount...
متن کاملPathway of nitrogen metabolism after fixation of 13N-labeled nitrogen gas by the cyanobacterium, Anabaena cylindrica.
Methods have been developed for identifying the pathway of assimilation of N2-derived nitrogen. The products of fixation of 13N-labeled nitrogen gas ([13N]N2), and the distribution of 13N within glutamine, were determined after short periods of labeling (approximately 1 to 120 s) and also in pulse-chase experiments. Ammonia, the amide nitrogen of glutamine, and the alpha-amino nitrogen of gluta...
